We share a literature review of commercially available SGLT2 antibodies evaluated by immunohistochemistry and western blotting using kidneys and hearts from WT and SGLT2 deficient mice and rats. Antibody specificity was also evaluated in human kidney (Hirose et al., Hypertension, 2026). The authors perform a head-to-head screen of a panel of eight commercial SGLT2 antibodies.
We have rated the antibody performance according to the pAbmAbs criteria, and we highly encourage other scientists to share their experiences.
The antibody HPA041603 showed specific immunostaining in kidney sections from WT mice and rats (Figure 1) and no staining in cardiac sections. No staining was observed in Sglt2-/- sections from mouse and rat, confirming the specificity. The specificity of HPA041603 was further confirmed by preincubation with its antigen peptide which abolished staining in human and rodent kidney sections (Figure 2A).
Western blotting with this antibody detected a distinct band of the expected size using mouse and rat kidney homogenates as samples. However, a weak band of similar size was also present in Sglt2-/- samples from mouse kidney but not from rat (Figure 4). Curiously, a band of lower molecular weight was observed in both WT and KO samples from mice but absent in the rat KO sample.
