Review of: sphingosine 1-phosphate receptor 1, ABclonal, A12935 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Sphingosine 1-phosphate receptor 1 (S1PR1) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 9 commercial research antibodies against human S1PR1.
The collection of antibodies was tested using SK-HEP-1 WT and S1PR1 KO cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) A12935 detected a band of appr. 48 kDa in WT cell lysate with a faint corresponding band observed in KO cell lysate. Furthermore, an intense band of higher molecular weight was observed in both genotypes along with bands of lower molecular weight.
A smear was detected following IP (2 µg Ab used), this smear was more intense compared to band in input extract (4% of starting material). A faint smear was also detected in immunodepleted extract (4% of total unbound fraction), showing that the antibody does not completely deplete the antigen from cell lysate. In IF (1:1000 ab dilution), comparable staining intensity was observed in both KO and WT cells.
