Our team performed a comprehensive review of the scientific literature to identify studies using antibodies against human retinoic acid receptor RXR-alpha, followed by extraction and evaluation of available data according to pAbmAbs rating criteria.
YCharOS conducted a systematic head-to-head comparison of ten commercially available antibodies against retinoic acid receptor RXR-alpha across multiple applications.
Antibodies were tested by WB using lysates (100 µg protein) from HCT116 WT cells and retinoic acid receptor RXR-alpha deficient HCT116 (RXRA KO) cells . These cell lines were also used for IF. In addition, antibodies were evaluated by IP using HCT116 WT cell lysate (1 mg protein). All experiments were performed according to YCharOS standard protocols (see reference).
In WB (1:500 antibody dilution), 21218-1-AP detected a band at the predicted size (~50 kDa) in HCT116 WT cell lysate, with no corresponding band in RXRA KO lysate. However, several background bands were observed, including a band in the KO lysate very close in size to the specific band, making detection of the specific signal dependent on effective band separation.
In IP, the antibody (1µg) clearly enriched retinoic acid receptor RXR-alpha upon precipitation with no remaining protein detected in the unbound fraction. Retinoic acid receptor RXR-alpha levels in the starting material, unbound fraction, and precipitate were assessed by WB using the retinoic acid receptor RXR-alpha-specific antibody NBP2-75653 (1:500).
In IF, the antibody (1:500 dilution) failed to produce a signal in WT and KO cells.
