YCharOS is here conducting a large head-to-head study comparing 8 commercial research antibodies against human Optineurin encoded by OPTN.
The collection of antibodies was tested using U2OS WT and OPTN KO cells in the applications WB (WT and KO cell lysates/35 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).
In Western blot (1:1000 antibody dilution), 70928 detected a strong band at approximately 66 kDa in WT cell lysate, with no corresponding band in KO cell lysate. Several faint bands of both higher and lower molecular weight were observed in KO lysate, along with a strong band at approximately 120 kDa present in both genotypes. A band was detected following IP (2 µg Ab used). This band was markedly more intense than the corresponding band in input extract (4% of starting material). No band was detected in immunodepleted extract (4% of total unbound fraction), showing that the antibody does completely deplete the antigen from the cell lysate. In IF (1:400 ab dilution), a more intense staining was observed in WT cells compared to KO cells showing no background.
