NLRP3, GeneTex, GTX106313

Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human NLRP3 and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 14 commercial research antibodies against human NLRP3.

The collection of antibodies was tested using THP-1 WT and NLRP3 KO cells in the applications WB (WT and KO cell lysates/20 µg protein), WB on culture medium (WT and KO culture medium/20 µg protein) and IP (WT cell lysate) according to YCharOS standard protocols (see reference).

In WB (1:500 ab dilution) on cell lysate GTX106313 detected multiple strong bands. A band of appr. 100 kDa was detected in WT cell lysate, with no corresponding band observed in KO cell lysate. In WB (1:500 ab dilution) on culture medium a band of appr. 100 kDa was detected in WT not observed in KO. A strong band of lower molecular weight was observed in both genotypes. A band was also detected following IP (2 µg Ab used). This band was markedly more intense than band in input extract (4% of starting material), a band was also detected in immunodepleted extract (4% of total unbound fraction), showing that the antibody does not completely deplete the antigen from the cell lysate.
