Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human glucocerebrosidase (GCase) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 24 commercial research antibodies against human glucocerebrosidase encoded by GBA1 gene.

The collection of antibodies was tested using HAP1 WT and GBA1 KO cells in the applications WB (WT and KO cell lysates/30 µg protein) and IP (WT cell lysate). The collection of antibodies was also tested using RPE-1cells in the application IF (WT and GBA1 KO cells) according to YCharOS standard protocols (see reference).

In WB (1:1000 ab dilution) MA5-32591 detected a faint smear of appr. 63 kDa in WT cell lysate not observed in KO cell lysate. An intense band of higher molecular weight was observed in both KO and WT cell lysate. No band was detected following IP (2 µg Ab used). In IF (1:1000 Ab dilution), WT cells showed a more intense staining compared to KO cells, also showing some background fluorescens.
