Review of: Glucocerebrosidase, ABclonal, A19057 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human glucocerebrosidase (GCase) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 24 commercial research antibodies against human glucocerebrosidase encoded by GBA1 gene.
The collection of antibodies was tested using HAP1 WT and GBA1 KO cells in the applications WB (WT and KO cell lysates/30 µg protein) and IP (WT cell lysate). The collection of antibodies was also tested using RPE-1cells in the application IF (WT and GBA1 KO cells) according to YCharOS standard protocols (see reference). Furthermore, A19057 was tested using NPC (WT and KO), HeLa (WT and KO) and U-87 MG (WT) cell lysates in the application WB (30 µg of protein).
In WB (1:1000 Ab dilution) A19057 detected a band of appr. 63 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. Furthermore, lysates were prepared from WT and GBA1 KOs in HAP1, neural progenitor cells (NPC) and HeLa as well as WT U-87 MG and RPE-1 and processed for western blot with A19057 (1:1000 Ab dilution). In NPC a band of appr. 63 kDa was observed with no corresponding band in KO. In HAP1 and HeLa a faint band of appr. 63 kDa was observed with no corresponding band observed in KO. U-87 MG and RPE-1 detected a band of appr. 63 kDa. No band was detected following IP (2 µg Ab used). In IF (1:500 ab dilution), comparable staining intensities were observed for both KO and WT cells.
