Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Casein kinase I isoform alpha and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 10 commercial research antibodies against human Casein kinase I isoform alpha encoded by CSNK1A1.

The collection of antibodies was tested using HCT 116 WT and CSNK1A1 KD cells in the applications WB (WT and KD cell lysates/50 µg protein), IP (WT cell lysate) and IF (WT and KD cells) according to YCharOS standard protocols (see reference).

In WB (1:500 Ab dilution) ARP76053 detected a faint band of appr. 39 kDa in WT cell lysate with no corresponding band in KD cell lysate. Several bands of higher molecular weight were observed for both genotypes and with stronger intensity. A band was also detected following IP (2 µg Ab used). This band was more intense than the corresponding band in input extract (4% of starting material). No band was detected in immunodepleted extract (4% of total unbound fraction), showing that the antibody does completely deplete the antigen from the cell lysate. In IF (1:250 Ab dilution), comparable staining intensity was observed for both genotypes.
