Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Calpain-2 catalytic subunit and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 13 commercial research antibodies against human Calpain-2 catalytic subunit encoded by CAPN2.
The collection of antibodies was tested using MDA-MB-231 WT and CAPN2 KO cells in the applications WB (WT and KO cell lysates/20 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference). Additionally, WB was carried out in HEK293T (WT and CAPN2 KO) for antibody ab126600.
In WB (1:5000 Ab dilution) of MDA-MB-231 (WT and CAPN2 KO) ab126600 detected a single band of appr. 74 kDa in WT lysate, with no band observed in KO lysate (figure 1A). In WB (1:5000 Ab dilution) of both MDA-MB-231 (WT and CAPN2 KO) and HEK293T (WT and CAPN2 KO), a single band of appr. 74 kDa was detected for both cell type lysates during long exposure time, with no corresponding band in KO cell lysates (figure 1B middle panel). During short exposure time (figure 1B top panel) an intense band was observed in MDA-MB-231 WT cell lysate, with no corresponding band I KO cell lysate. In cell lysate from HEK293T ab126600 detected a faint band with no corresponding band in KO cell lysate. No band was observed in the following IP (1 µg Ab used). ). In IF (1:1000 Ab dilution), comparable staining intensities were observed in both KO and WT cells.
