Our team performed a comprehensive review of the scientific literature to identify studies using antibodies against human TYRO protein tyrosine kinase-binding protein, followed by extraction and evaluation of available data according to pAbmAbs rating criteria.
YCharOS conducted a systematic head-to-head comparison of 9 commercially available antibodies against TYRO protein tyrosine kinase-binding protein across multiple applications.
Antibodies were tested by WB using lysates (100 µg protein) from THP-1 WT (ab271147) and TYRO protein tyrosine kinase-binding protein deficient (TYROBP KO) (ab273726) cells. These cell lines were also used for IF. In addition, antibodies were evaluated by IP using THP-1 WT cell lysate (1 mg protein). All experiments were performed according to YCharOS standard protocols (see reference).
In WB (1:1000 antibody dilution), 12492 detected a single band in the THP-1 WT lysate below the 11 kDa marker, with no corresponding band in TYROBP KO cells, supporting specificity. Although this is lower than the expected size (~12 kDa), the signal is consistent with the expected band when compared across antibodies.
In IP, the antibody (1 µg) clearly enriched TYRO protein tyrosine kinase-binding protein upon precipitation, with little detectable target protein remaining in the unbound fraction. TYRO protein tyrosine kinase-binding protein levels in the starting material, unbound fraction, and precipitate were assessed by WB using the TYRO protein tyrosine kinase-binding protein-specific antibody 12492 from Cell Signaling Technology (1:1000).
In IF, the antibody (1:40 dilution) produced a clear TYRO protein tyrosine kinase-binding protein-specific staining distributed throughout THP-1 WT cells, with little background in TYROBP KO cells
