Used on (frozen) human PBMCs in a 10 color flow panel on LRS-Fortessa.
Recommended use: 10µL/100µL. Titrated as single stain and found 0.625µL to be sufficient. However, for it to work perfectly in the 10-color panel I had to increase the volume and use 2µL.
Can easily distinguish the CD56-,CD56dim, and CD56 bright (TLR2- NK , T and B cells)
On a plot against APC-TLR2+, I had to use FMO to find the CD56 negative gate,.
Samples are blocked using 10% human AB serum and stained fifty/fifty in PBS with 1% BSA and Brilliant Stain Buffer (BD horizon), and fixed in 0.9% formaldehyde. Compensated on OneComp eBeads (Thermofisher Scientific)
