This antibody works really well to detect activation of TrkB at site Y706/707. Clear bands were detected in wild-type primary cortical neuronal lysates treated with 1nM BDNF. ECL Prime system was used for imaging.

This is one of the best antibodies for westernblotting differences in phosphorylation after stimulation that i have ever used. I used it succesfully on both human and mouse TrkB with great differences between stimulated and unstimulated cells.

This antibody was tested on PFA-fixed cultured hippocampal neurons which had either been stimulated with 25ng/ml BDNF or similar voulme of sterile PBS for 10 minutes.
high background was observed and no difference was seen between neurons with or without BDNF stimulation. when using a dilution of 1:1000.
This pTrkA/B antibody from CST is excellent, #4621. I have been working with phospho-Trk antibodies for (too) long and this is one of the best ones. Only problem is that is does not differentiate between different Trk family members. Nicely detects BDNF (and NGF) induced TrkB (TrkA) phosphorylation in Trk expressing cells (cell lines, primary neurons) in western. Also works beautifully with brain tissue lysates (e.g. antidepressant-drug induced Trk phosphorylation). Highly recommend.

We use it for detecting trans-phosphorylated TrkB upon BDNF stimulation of primary mouse hippocampal neurons or 293 cells stably expressing human TrkB.
We get a very nice BDNF concentration-dependent trans-phosphorylation of TrkB. This is a quick and efficient way to assess TrkB activation in cultures by Western blotting.
WB dilution 1:1000
Diluted antibody can be frozen and reused.
