This antibody yielded a clear band of pPLCgamma from BDNF-stimulated mice primary cortical neurons without background. In lanes with lysate from unstimulated neurons the bands was weak or undetectable.
I have used kinexus lysis buffer for the lysates and exposed for 15-25 minutes utilizing ECL™ Prime (Amersham).
This antibody worked well giving clear bands of p-PLCgamma from primary hippocampal neurons. These were lysed in kinexus lysis buffer. The antibody did, however, NOT work using TNE lysis buffer.

I was able to succesfully get a signal using this antibody with no band in the unstimulated lane. It is however somewhat weak. (see picture)
Konc.: 1/1000
Exposure-time: 25 min
lane-load: 50 ug

I used this in a dilution of 1:1000 as advised by the manufacturer for western blotting. The samples were murine cell lysates from a primary hippocampal neuron culture. I was not able to make the antibody yield a succesful blot.

1:1000 from manufacturer. Needs to be exposed for at least 10 min. Demands a protein-load over 30ug/lane. Gives a signal by BDNF-stimulation of SY5Y cells transfected with TrkB, which is not observed in control lanes.
