Dilution: 1:100 in PBS or in blocking/permeabilization buffer (buffer containing 5% normal goat serum (NGS) and 0.1-0.3% Triton-X 100 in 1xPBS)

This AB is suppose to detect pro-caspase3 as well as celaved caspase-3. However, I only maneged to detect pro-caspase3 in primary blood purified CD4+ T cells ( treated with stausporine). One fine band for the pro-caspase3, but no band for the celeaved active capse3, therefor only 2 stars. Remeber to block in skim milk.

There is a nice increase in activated caspase-3 staining in the DRG of the injured side. And also in the fiber in Schwann cells closed to the site of injury. This increase is not seen in the uninjured side.
The antibody only gets three stars because there appears to be some batch variations for this antibody. One of the batches we have acquired may show cross reactivity with uncleaved caspase-3.
